High density of ‘spiky’ excrescences covering the surface of an erythrocyte infected with Plasmodium malariae

چکیده
This Atomic Force Microscope image shows the surface of a Plasmodium malariae trophozoite-infected red blood cell isolated from a Burmese patient suffering from quartan malaria (top). Of note, the P. malariae-infected cell (bottom left) is covered with dense ‘spike-like’ excrescences (mean height: 7Æ59 nm; mean diameter: 52Æ95 nm), which are morphologically distinct from the larger, more rounded ‘knob’ structures found on a Plasmodium falciparum-infected red cell (mean height: 19Æ65 nm; mean diameter: 96Æ64 nm) (bottom right). The ‘knobs’ on red cells containing mature asexual forms of P. falciparum assist the infected cells to bind/sequester to the vascular endothelium under shear flow conditions and thus avoid splenic clearance. The function of P. malariae spikes (which we have observed on every sexual and asexual stage examined in isolates from Indonesia and Thailand) is not known. The sample from which the P. malariae pictured was isolated (Mae Sod, Tak, Thailand) had the diagnosis confirmed by polymerase chain reaction. This picture is artificially coloured. The horizontal black scale bars represent 1 lm. The two lower images are identical in scale and orientation. Ang Li Bruce Russell Laurent Renia Usa Lek-Uthai Francois Nosten Chwee T. Characterization of a modified red cell membrane protein expressed on erythrocytes infected with the human malaria parasite Plasmodium falciparum: possible role as a cytoadherent mediating protein
Infections with the human malaria Plasmodium falciparum are characterized by the retention of parasitized erythrocytes in tissue capillaries and venules. Erythrocytes containing trophozoites and schizonts attach to the endothelial cells that line these vessels by means of structurally identifiable excrescences present on the surface of the infected cell. Such excrescences, commonly called knobs...

We examined the ultrastructure of Plasmodium malariae-infected erythrocytes in peripheral blood and tissue biopsies of the liver, stomach, and duodenum from three patients infected with P. malariae. Ultrastructural features of P. malariae-infected erythrocytes in peripheral blood appear similar to those described previously. The surface membranes of P. malariae-infected erythrocytes had numerou...
متن کاملClinical Pharmacology of the Antimalarial Chloroquine in Children and Their Mothers
Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are the parasites that infect humans. Plasmodium falciparum and Plasmodium vivax cause most of the malarial infections worldwide. Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are susceptible to chloroquine. Chloroquine was the world's most widely used antim...
متن کاملPlasmodium falciparum-Infected Erythrocyte Knob Density Is Linked to the PfEMP1 Variant Expressed
UNLABELLED Members of the clonally variant Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) family mediate adhesion of infected erythrocytes (IEs) to vascular receptors. PfEMP1 expression is normally confined to nanoscale knob protrusions on the IE surface membrane. To investigate the relationship between the densities of these IE surface knobs and the PfEMP1 variant expressed, we ...
متن کاملMerozoite surface protein-1 genetic diversity in Plasmodium malariae and Plasmodium brasilianum from Brazil
BACKGROUND The merozoite surface protein 1 (MSP1) gene encodes the major surface antigen of invasive forms of the Plasmodium erythrocytic stages and is considered a candidate vaccine antigen against malaria. Due to its polymorphisms, MSP1 is also useful for strain discrimination and consists of a good genetic marker. Sequence diversity in MSP1 has been analyzed in field isolates of three human ...
